Protective Effect of Ethyl Acetate Fraction from Domestic Walnut (Juglans regia) against PM2.5-induced Inflammation and Apoptosis

Abstract

This study examined the protective effect of the ethyl acetate fraction from domestic walnut (Juglans regia) as a Gimcheon 1 ho (EFGC) cultivar on particulate matter (PM)2.5-induced cytotoxicity in hippocampal HT22 cells. EFGC had the highest total phenolic contents, total flavonoid contents, and total tannin contents compared to other fractions. In addition, EFGC showed significant 3-ethylbenzothiazoline-6-sulfonic acid (ABTS), and 1,1-diphenyl-2-picrylhydrazl (DPPH) radical scavenging activities and ferric reducing/antioxidant power (FRAP). The EFGC presented an inhibitory effect of malondialdehyde (MDA) production and acetylcholinesterase (AChE) activity. The EFGC effectively decreased intracellular reactive oxygen species (ROS) contents and cellular death on hippocampal cells against PM2.5 cytotoxicity. The EFGC improved inflammatory reaction and apoptosis by regulating protein expression, such as BCl-2, p-JNK, BAX, caspase-1, and p-NF-¥êB.
